PERCOBAAN 3

“ANALISA PIGMEN TUMBUHAN DENGAN MENGGUNAKAN KROMATOGRAFI KERTAS DAN SPEKTROSKOPI UV-VIS”

A.    Tujuan Percobaan

1.      Memisahkan zat warna dari ekstrak tumbuhan menggunakan kromatografi kertas.

2.      Menentukan panjang gelombang maksimum yang diserap tiap zat warna yang diperoleh.

3.      Menyarankan jenis dan struktur molekul klorofil dari data spektrum UV-Vis yang diberikan instrumentasi spektroskopi UV-Vis.

B.     Prinsip Percobaan

Memisahkan komponen zat warna dari ekstrak tumbuhan berdasarkan distribusinya terhadap fase diam (air yang terikat pada kertas) dengan fase gerak (etanol) yang sesuai dengan tingkat kepolarannya sehingga diperoleh kromatogram. Kemudian kromatogram yang diperoleh dipisahkan dan dilarutkan dengan pelarut etanol untuk di uji jenis dan struktur molekul klorofil dengan menggunakan spektroskofi UV-Vis berdasarkan penyerapan energi radiasi elektromagnetik dari sinar UV-Vis dengan energi tertentu, sehingga elektron-elektron dalam larutan  sampel mengalami transisi elektronik dan kemudian elektron tersebut kembali ke keadaan dasar dengan panjang gelombang tertentu yang ditangkap oleh detektor dan ditampilkan dalam bentuk spectrum pada layar komputer.

C.    Dasar Teori

1.      Kromatografi kertas

Kromatografi adalah suatu teknik untuk memisahkan campuran menjadi komponennya dengan bantuan perbedaan sifat fisik masing – masing komponen yang berdasarkan partisi antara cuplikan dengan fasa gerak dan fasa diam.

Molekul yang terlarut dalam fase gerak, akan melewati kolom yang merupakan fase diam.Molekul yang memiliki ikatan yang kuat dengan kolom akan cenderung bergerak lebih lambat dibanding molekul yang berikatan lemah.Dengan ini, berbagai macam tipe molekul dapat dipisahkan berdasarkan pergerakan pada kolom.

Komponen utama kromatografi adalah fasa stationer dan fasa mobil dan kromatografi dibagi menjadi beberapa jenis bergantung pada jenis fasa mobil dan mekanisme pemisahannya, seperti ditunjukkan di Tabel 1.1.

Kromatografi  kertas merupakan salah satu metode pemisahan berdasarkan distribusi suatu senyawa pada dua fasa yaitu fasa diam dan fasa gerak. Pada kromatografi kertas, fase diam adalah air yang terikat pada kertas dan fase geraknya adalah pelarut organik.  Pada kromatografi kertas penampakan noda dapat dilihat hanya dengan disemprot dengan pereaksi spesifik, seperti: dragendorff, serium sulfat, ninhidrin, dan FeCl₃. Analisis kualitatif menggunakan kromatografi kertas dilakukan dengan cara membandingkan harga relative response factor (Rf). Nilai Rf identik dengan time retention (tR) atau volume retention (VR).
